Autoimmune diseases are a group of diverse diseases with a common cause in which the immune system responds to self-antigens, leading to damage or dysfunction of tissues. Biomarkers are measurable substances in the body associated with the intensity of a disease or other physiological states. They can be used in the diagnosis, prognosis, and treatment of different diseases. The summer internship project aims to:

    • Use clinical samples of autoimmune diseases with respective control group samples

    • Screen for miRNA, mRNA, and Protein biomarkers using high-plex multi-omics spatial transcriptomics platforms to identify disease-specific or common autoimmune biomarkers

    • Use the NanoString nCounter platform to identify miRNA biomarker candidates in clinical serum samples of autoimmune disease patients. This platform uses a unique barcode system and can directly detect up to 799 target genes in a single sample without amplification. FFPE or freshly frozen tissue samples will be used for the spatial transcriptomic analysis. By unraveling the transcriptomic biomarkers at miRNA, RNA, and protein levels, the project aims to understand the disease-specific biomarkers of autoimmune diseases at the transcription and translational levels.

    • The intern will be involved in

    • Sample processing, reagent preparation, reaction setup, instrumentation learning, and data analysis for the nCounter and spatial transcriptomic platforms

    Internship Development Opportunities:

    • After the internship, the intern is expected to be proficient in the high-plex multi-omic assay workflow, understand hypothesis-driven experiment design, and be able to share findings at Takeda's inter departmental meetings.
